The lipid composition and membrane fluidity of Dictyostelium discoideum plasma membranes at various stages during differentiation.

چکیده
There are only minor changes in the amounts of the major lipid constituents of Dictyostelium discoideum plasma membranes during the early stages in the differentiation of this organism. By the time cells reach the pseudoplasmodial stage of development there are small increases in the amounts of phosphatidylinositol, phosphatidylglycerol, and lysophosphatidylethanolamine, and small decreases in the amounts of phosphatidylethanolamine and its plasmalogen form. There is also a slight decrease in the total amount of sterol in the plasma membrane during the transition from aggregation to pseudoplasmodium formation. However, no significant change in membrane fluidity as determined by electron paramagnetic resonance (EPR) accompanies these minor changes in lipid composition. It can be concluded that the establishment of cell-cell interaction in D. discoideum does not involve gross changes in plasma membrane fluidity or lipid composition. It was found that the plasmalogen form of phosphatidylethanolamine is a major phospholipid constituent in D. discoideum, and that this species is somewhat enriched in the plasma membrane.-Weeks, G., and F. Preparation of Dictyostelium discoideum for electron microscopy.
Dictyostelium discoideum grew and differentiated normally on cellulose dialysis membranes placed on agar plates. This permitted the examination of cells at various stages of development, including cells during aggregation. Cells were preserved well and fixed for electron microscopy by brief exposure to acrolein vapore followed by immersion in glutaraldehyde solution.
متن کاملA membrane cytoskeleton from Dictyostelium discoideum. I. Identification and partial characterization of an actin-binding activity
Dictyostelium discoideum plasma membranes isolated by each of three procedures bind F-actin. The interactions between these membranes and actin are examined by a novel application of falling ball viscometry. Treating the membranes as multivalent actin-binding particles analogous to divalent actin-gelation factors, we observe large increases in viscosity (actin cross-linking) when membranes of d...

Changes in phospholipids of plasma r .embranes and total particulate membranes of the cellular slime mold Dictyostelium discoideum were analyzed during development. The predominant classes of phospholipids in the plasma membrane were phosphatidylethanolamine and phosphatidylcholine, constituting approximately 50 and 25 molar per cent of phospholipid phosphorus, respectively. The relative amount...

Studies were carried out on microsomes isolated from the highly differentiated (slow-growing) Morris hepatoma 9618A, on microsomes and plasma membranes from the poorly differentiated (fast-growing) Morris hepatoma 3924A, and rat liver used as control. The lipid composition (phospholipid and cholesterol content, degree of fatty acid unsaturation) and peroxidation of such membranes has been corre...
متن کاملIsolation of concanavalin a caps during various stages of formation and their association with actin and myosin
Regions of plasma membrane of dictyostelium discoideum amoebae that contain concanavalin A (Con A)-receptor complexes are more resistant to disruption by Triton X-100. This resistance makes possible the isolation of Con A-associated membrane fragments in sufficient quantity and homogeneity to permit the direct biochemical and ultrastructural study of receptor-cytoskeletal interactions across th...
